Well, the title pretty much says it all... I believe there may have been another post, but that was mostly just to name off all the cool combinations, but which one(s) is/are your favorite?
Mine is definitely Pihrannha + Plane

Heart + Coin + Anything else which goes (OSF/Mochi) Shy Guy + Heart + Car (OSF/Mochi) Boo + Coin + Heart — everything else is used for sustaining (Steinway) Boo + Star (Real)
I could bend the combinations a lot more on FL (my most used combination being Electronica AND Real Stars), but on MPC, I often use the above.

For a more powerful bass, I combine plane, dog and heart. For a more powerful melody, I combine cat, flower and gameboy. For a more powerful harmony, I combine mario and star. For more powerful backing chords, I combine piranha plant, shy guy and car. For more powerful percussion, I combine mushroom, boat, yoshi, face and pig.
You know what that leaves? Coin and boo. Me no likey.

One combo I´ve used since forever is the percussion (Mushroom, Boat) along with "Low-A-Glitched" Yoshis, Faces, Pigs and/or Dogs. Such combo gives a weird cartoonie beatbox effect to the whole percussion. Some easy examples can be found in the funky part of the Astro Chase (before the loop point), in the 1st Community Composition right before Levus´ part and at the beginning of the 5th Community Composition. Also, many of mario7mario3 songs use that combo in certain way.

I abuse coin and boo way too much. But I love it because of the great ambiance it gives to songs.

1. Low mushroom + Heart = THE KITCHEN
Add a plane / piranha plant / gameboy / dog and you've got yourself a fricking rock band
2. Flower + Dog

I like Plane + Piranha Plant and Car + Flower because they each sound like one instrument instead of two. I also tend to overuse Car + Piranha Plant and Heart + Piranha Plant.

I always enjoy the heart and plane combo on original soundfont. It sounds so folky.

Here is my breakdown of my instrument selection/arranging style with Mario Paint.
- for harmonic thirds, I use Flower's (upper) and Gameboy's (lower). - the planes are my favorite choice for harmony/broken chord structures. - inverted octave with the hearts (often used to accommodate note placement where melody dominates) - the octave with Mario (upper) and the Swan (lower) = incredible timbre!! - I often switch instruments between phrases of the melody, usually to mask the octave placement of the pitches and to generate more appreciation for the various melodic instruments Mario Paint has to offer. - percussion = Mushroom (low B or C) and boats (middle line B, or A if in use) - Cars are my least favorite (unless melody calls for slow, legato passages). - Dogs, I find, are more appropriate usually in moments where the melody is absent, usually on the tonic or dominant. - Cats have become a meme of sorts in my arrangements ("gotta have at least one!"), though their placement/appropriateness can be tricky. - Yoshi's I find work best if placed on a pitch matching the tempo of the work. I fancy the low B placement, where the second part of the sound is often canceled by the following notes. - Swans are used for rhythmic accents, or brief melodic phrasing, usually in combination with another voice for embellishment. - The face is one of those sounds that... yeah, the planets have to align in such a way to ever find an acceptable placement for this unique instrument. I often place them in moments of complete silence, and on a particular pitch that matches the tempo of the piece. - Pig... useless in most cases. - Stars I find to be useful to extend melodic passages an octave higher than Mario's. - my arrangements try to stay true to the original melody, and avoid any that require extensive modification due to chromaticisms. I always love to add my own original material (usually the harmony), and retain familiarity with the bass line.
